Due to failures of infection after mechanical inoculation of begomovirus on tomato (Lycopersicon esculentum Mill.) plants, an efficient and reliable begomovirus inoculation method was developed using the whitefly vector (Bemisia tabaci). Virus acquisition was carried out using a 'Tube-cage', made of a polypropylene tube, containing one to two day-old whiteflies and a tomato plant apex infected with a begomovirus isolate. ABSTRACT: Insect-transmitted viruses cause some of the most damaging and economically important diseases of crop plants, especially in tropical and subtropical regions. The geminiviruses, a group of single-stranded DNA viruses with unique twined icosahedral virus particles, are responsible for many of these diseases. Of particular importance are the geminiviruses transmitted by whiteflies (Bemisia tabaci), which are in the genus Begomovirus. This is the largest genus of plant viruses (in terms of number of species), and some are responsible for many devastating diseases in vegetable and fiber crops throughout the world. A mancha-bacteriana e a begomovirose são exemplos de doenças importantes em tomate para processamento industrial no Brasil em que a disponibilização de cultivares com resistência é extremamente desejável. No presente trabalho foram conduzidos três ensaios (2010 e 2011) para avaliar híbridos comerciais e experimentais de tomate para processamento industrial em campo, com ocorrência de begomovirose (ensaios 1 e 3) e de mancha-bacteriana (ensaios 2 e 3). O Nordeste é a principal região produtora de melão do País, contribuindo com mais de 90% da produção nacional. Entretanto, o "amarelão do meloeiro", doença associada à presença de um novo vírus, o Melon yellowing-associated virus (MYaV), vem afetando a cultura desde 1999. Este trabalho teve como objetivo detectar a ocorrência do MYaV em meloeiros exibindo sintomas suspeitos da doença nos estados que são os maiores produtores da Região Nordeste. Em novembro de 2007, plantios comerciais de melão foram inspecionados para avaliar a ocorrência do "amarelão". The number of tomato-infecting begomoviruses has increased in Brazil after the introduction of the polyphagous vector Bemisia tabaci biotype B. The Ty-1 locus, introgressed from Lycopersicon chilense, controls tolerance to species of the monopartite Tomato yellow leaf curl virus (TYLCV) complex in Europe and the Middle East. However, little information is available about the Ty-1 effectiveness against species of the bipartite Begomovirus complex occurring in Brazil.
